摘要:溴酚蓝在作为检测牛胰岛素淀粉样纤维探针和作为牛胰岛素淀粉样纤维抑制剂方面的应用,属于生物医学技术领域。荧光光谱图显示溴酚蓝加到牛胰岛素淀粉样纤维溶液后,荧光强度是牛胰岛素溶液与溴酚蓝混合后的三倍多,由于溴酚蓝与牛胰岛素和牛胰岛素淀粉样纤维结合后荧光强度的明显差异性,表明溴酚蓝可以作为探针准确检测牛胰岛素淀粉样纤维。用溴酚蓝追踪监测牛胰岛素淀粉样纤维的形成过程,第一阶段荧光强度变化较为缓慢,第二阶段荧光强度迅速增强,第三阶段荧光强度变为较为平稳。在制备牛胰岛素淀粉样的过程中,加入溴酚蓝后,圆二色谱图显示牛胰岛素的二级结构没有出现应该有的变化,即溴酚蓝的加入成功抑制了牛胰岛素淀粉样纤维的形成。 | |
Abstract: The invention relates to an application of bromophenol blue as a probe for detecting bovine insulin amyloid fiber and an inhibitor for the bovine insulin amyloid fiber. A fluorescence spectrogram displays that bromophenol blue is added in a bovine insulin amyloid fiber solution, the fluorescence intensity is three times of that of a mixture of the bovine insulin solution and bromophenol blue, andbecause fluorescence intensity has obvious difference after the bromophenol blue and bovine insulin and bovine insulin amyloid fiber are combined, the bromophenol blue can be used as the probe for accurately detecting the bovine insulin amyloid fiber. The bromophenol blue is used for tracking and monitoring a formation process of the bovine insulin amyloid fiber, change of the fluorescence intensity of a first phase is slow, the fluorescence intensity of a second phase is rapidly enhanced, and the fluorescence intensity of a third phase is stable. During a bovine insulin amyloid fiber preparation process, after bromophenol blue is added, a circular dichroism spectrum displays that the change of the secondary structure of the bovine insulin does not appear that should be, so that the addition of the bromophenol blue successfully inhibits the formation of the bovine insulin amyloid fiber. | |
