摘要:本发明公开了一种PCV2病毒样颗粒及其制备方法和裂解及VLP组装缓冲液。基于自主优化设计,并人工合成适合在原核表达系统中高效表达的PCV2核衣壳蛋白基因,通过大肠杆菌原核表达系统表达PCV2核衣壳蛋白的全长基因序列,并利用其可溶性核衣壳蛋白在特定条件下高效自体组装成了病毒样颗粒。本发明创新点主要在于采用不同于常规的真核表达系统获取VLPs方法,而是利用原核表达系统获取PCV2VLPs,本方法成本低廉、简单高效,适合大规模产业化应用;其中还应用到独创的既可以促进菌体裂解又能适合VLPs自体组装的双功能于一体的缓冲液配方;另外发明所获得的PCV2病毒样颗粒与野生型病毒外形相似度高,并且免疫原性好,可以用于研发具有应用潜力的猪圆环病毒的亚单位疫苗及药物传递载体。 |

Abstract: The invention discloses PCV2 virus-like particles as well as a preparation method thereof and splitting and VLP assembly buffer liquor. Based on an autonomous optimization design, a PCV2 nucleocapsid protein gene which is suitable for efficiently expressing in a prokaryotic expression system is artificially synthesized, a full-length gene sequence of the PCV2 nucleocapsid protein gene is expressed by an escherichia coli prokaryotic expression system, and the virus-like particles are efficiently and autonomously assembled by utilizing soluble nucleocapsid protein of the full-length gene sequence under a special condition. An innovation point of the invention is that PCV2VLPs are obtained by utilizing the prokaryotic expression system instead of adopting the conventional method for obtaining VLPs through an eukaryotic expression system; the method is low in cost, simple and efficient, and suitable for large-scale industrial application; moreover, an innovative buffer liquor formula integrating double functions, which not only can promote thallus splitting, but also can be suitable for self-assembling of VLPs, is also applied; besides, the PCV2 virus-like particles obtained in the invention are very highly similar with wild type virus in outline and good in immunogenicity, and can be applied to developing a subunit vaccine and a drug delivery carrier with utilization potentiality of porcine circovirus. | |

主权项 | 一种PCV2病毒样颗粒的制备方法,其特征在于,依次包括以下步骤:1)选取利用密码子的兼并性,人工合成的适合于原核表达的编码PCV2核衣壳蛋白全长基因;2)将基因序列与表达质粒载体重组得到的重组质粒转化大肠杆菌,然后加入IPTG,使得体系中其浓度为0.1?1mM,并在25?37℃进行诱导表达;3)将诱导表达PCV2核衣壳蛋白时获取的细菌沉淀用以下配方的裂解及VLP组装缓冲液重悬,超声处理,离心收集上清,树脂吸附目的蛋白,然后从树脂上洗脱目的蛋白;所述的裂解及VLP组装缓冲液配方为:0.1M?NaH2PO4·2H2O、0.1M?Na2HPO4、20mM?Imidazole、10mM?Tris?base、300mM?NaCl、50mM?KCl、2mM?MgCl2、0.1M?Ammonium?citrate、2%Glycerine、0.5%Triton?X?100、5mMβ?Me、0.1mM?PMSF、1U/mL?Leupeptin?Protease?inhibitor;其中Triton?X?100、β?Me、PMSF、Leupeptin?Protease?inhibitor现用现加,缓冲液pH8.0,余量为纯水。 | ||||||||||||||||||
